Rijn, 31-3-02-04 11 Reptilia, testudines, Gopherus agassizzi, infection, bacteria, Mycoplasma agassizii., URTD A long term study is currently being conducted on upper respiratory tract disease (URTD) in the desert tortoise, Gopherus agassizii to determine the etiologic agent and to elucidate the mechanism of pathogenesis. Clinically healthy desert tortoises and those with URTD were sacrificed and nasal cavity tissue was prepared for electron microscopy. Examination revealed all ill and the majority of the clinically healthy animals possessed pathological changes consistent with URTD. Inflammatory cells were observed near the luminal surface where mycoplasma's were attached to the apical membrane of both secretory and ciliated cells. Cytoadhesion was usually by generalized attachment to the cell surface. Mycoplasmal colonization of the nasal cavity appeared to be regional, though the lesions produced did not. Hyperplasia and hypertrophy, widening of the intercellular spaces due to fluid accumulation and infiltration of immune and inflammatory cells were moderate to extreme. Total loss of the normal architecture, accompanied by basal cell metaplasia, occurred in some areas. The findings reflect the chronic nature of URTD and are comparable to respiratory mycoplasmosis described in rodents. Experiments to identify and differentiate possible direct cytopathic effects from injuries that are the result of the host inflammatory response are now in progress. Tortoise tracheal ring and fibroblast cell cultures are employed to examine the potential cytopathic effects of different isolates of Mycoplasma agassizii. Tracheal tissue from the gopher tortoise, Gopherus polyphemus, is also being used for comparison. A difference in target cell specificity was detected between two isolates that cause similar pathological changes in free-ranging populations. These same differences were observed in the gopher tortoise which has been shown to be infected with the same species of mycoplasma in the wild. Using a general linear modeling program, we compared mean values for these parameters with mean values of a group afflicted with green turtle fibropapillomas (GTFP). Turtles of similar size classes from both groups were collected under the same conditions in the same study area and season at the same time of the day. Corticosterone, hematological, and enzymatic responses to acute and chronic stress were characterized for each group at four different sampling periods: 0 h (within 2 min of capture), 1 h, 3-4 h, and 24 h postcapture. On the basis of the differences identified between groups and times within a group, we conclude that turtles with GTFP are chronically stressed and immunosuppressed. Eosinophilic granulation of the nucleus and intra-nuclear inclusion bodies appeared between 30-60 h pi. Small round and ring-shaped (type I), circular, semicircular or rod-shaped (type II) and large, sharp, round (type III) inclusion bodies were formed. Typical crystalline aggregates of virions appeared at the periphery of the nucleus 48-60 h pi. The release of virions into the cytoplasm was followed by lysis of the infected cells. Measurements of egg size and shape, as well as position of the clutch within the coelomic cavity, were taken from diag nostic radiographs of 20 female Cuban rock iguanas, Cyclura nubila, 81 to 18 days prior to laying. Combined with data on maternal body size, these variables were entered into multiple regression models to predict clutch size and timing of egg laying. The model for clutch size was accurate to 0.53 +,- 0.08 eggs, while the model for oviposition date was accurate to 6.22 +,- 0.81 days. To study the proliferation of and protein synthesis in fat cells, tritiated thymidine and proline were injected into lizards after 3 weeks of tail regeneration and the tracers were revealed by optical and electron microscopic autoradiography. After 3 weeks of tail regeneration, adipocytes are rarely seen in the apical regions of the regenerating tail and most of the preadipose cells are visible in the submuscular connective tissue in the proximal regions. Fat cells begin to proliferate and accumulate visible lipid droplets in the proximal submuscular connective tissue after about 3 weeks of tail regeneration. The kinetics of [H-3]thymidine uptake after progressive postinjection periods suggest that fat tissue mass increases mostly as as result of fat cell hypertrophy, with only limited cell multiplication. During the initial stages of their differentiation, lipoblasts also take up [H-3]proline, but the ergastoplasm and Golgi apparatus become reduced during the process of fat accumulation. At the apex of the regenerating tail, groups of 4-8 myoblasts of the promuscle aggregates fused to produce bundles of myotubes whose multiple labelled and unlabelled nuclei appeared to be distributed at random. The formation of the first myotubes and their growth is responsible for the formation of the myotome primordia and their separation from the intermuscular connective myosepta. More nuclei were added with the lengthening of the myotubes-up to 14-18 nuclei in the oldest proximal myotubes. At 4-5 h after injection labelled nuclei were found outside the myotubes while at 2-6 d after injection many labelled nuclei were observed in the myotubes, particularly near the two ends of the myotubular sarcoplasm contacting the myoseptum. This change from the initial distribution suggests that the growth of the myotubes takes place mostly at their terminals. There is an apparent correlation between the number of nuclei and the final length of the myotubes and myotome. The insertion of fibres with a similar number of nuclei and lengths into the pinnated connective myoseptum of the original musculature, the autotomy plane, probably determines the wave-like shape of the muscles within the regenerated myotomes. Although juveniles and adults (>0.6 m) may be sexed effectively, smaller alligators are reportedly difficult to sex. We developed a method for sexing young alligators by inspection of the genitalia and quantified effects of incubation temperature on genital morphology. We predicted the sex of a hatchling by examining its genitalia within the cloaca. Our predictions were verified by inspecting the gonads macroscopically and histologically. The male clitero-penis typically is red, round, and large (mean length = 4.1 mm) in comparison with the female organ (mean length = 2.8 mm), which is white and smaller. Sex identification by cloacal examination was highly reliable (accuracy greater than or equal to 96%). Clitero-penis dimensions of hatchlings incubated at 29, 31, 32, 33, and 34 C increased significantly with increasing incubation temperature, both within and between sexes. Based on differences apparent in hatchlings, incubation temperature seems to affect genital differentiation during development in a graded fashion. During posthatching growth, size increased rapidly in males versus females; based on previous studies, we think that these changes can be attributed to ontogenetic changes in the levels of circulating sex steriods. Observable size differences in the genitalia of males versus females permit an accurate, noninvasive diagnosis of sex in young alligators. Sex identification by this method will facilitate experimental study, as well as the management of wild and captive alligators. Rijn, 31-3-02-04 17 Reptilia, sauria, Lacerta schreiberi, infection, virus, erythrocytic virus Erythrocytic viruses were detected in the Iberian endemic lizard Lacerta schreiberi (Bedriaga, 1878) from Serra da Estrela and Serra do GerÍs, Portugal, and Serra de Bejar, Spain. Intraerythrocytic cytoplasmic viral inclusions were found during the spring and summer of 1994 in a high percentage of captured animals. The viruses are large (~220 nm) icosahedral particles with a two-layered core, infecting up to 100% of the erythrocytes. Six animals were inoculated i.p. with 0.1 ml of infected blood from a naturally infected animal from Serra da Estrela. Blood smears were made at 1 week intervals or less. For TEM, blood from a toe tip was collected into a capillary tube and processed following standard techniques. Organ fragments from sacrificed animals were processed for TEM and histological examination. Three of the inoculated animals developed erythrocytic infection in 2 to 3 weeks p.i.. The percentage of infected erythrocytes increased to 100% within a week and virions were produced within 1 month. The infection progressed towards elimination of the erythrocytic inclusions. The animals were active throughout the infection. The remaining three animals showed progressive and severe prostration from 2-3 weeks p.i. onwards. Two of these animals were sacrificed after the onset of symptoms and the third died. Internal bleeding was present in the peritoneal cavity and histological examination disclosed hepatitis with necrosis. TEM did not show so far virus particles in liver lesions. Few infected erythrocytes were seen in these animals before or at the onset of symptoms and they did not increase in number with time. Our results indicate that the L. schreiberi erythrocytic virus may infect inoculated animals. Further experiments and study of captive animals are necessary to rule out the possible activation of endogenous viruses in the laboratory. The course of the erythrocytic infection follows closely the pattern described by some of us(1) for other reptile erythrocytic viruses and seems largely asymptomatic. However, half of the animals did not develop blood infection, but instead a severe disease of uncertain etiology. It is presently unclear if this disease is related to the virus, since no replicating viruses were yet found in the affected organs. It is also unknown if it occurs in field populations since no similarly diseased animals were ever captured in the field. Food habits Herpetological Review 26 4 206 Sistrurus catenatus Journal Article Arad, Z. 1995 Physiological responses to increasing ambient temperature in three ecologically different, congeneric lizards (Gekkoninae: Ptyodactylus) Comparative Biochemistry and Physiology A - Physiology 112 2 305-311 Reptilia, sauria, gecko, Ptyodactylus, physiology, thermoregulation, oxygen consumption, diurnal, nocturnal Interspecific differences in the responses to increasing ambient temperature (up to 40 degrees C) were studied in three species of Ptyodactylus that differ in their distribution in Israel, in their daily activity pattern and in their preferred body temperatures: the northern, diurnal P, puiseuxi, the southern, nocturnal P, hasselquistii and the diurno-nocturnal P. guttatus, The diurnal P, puiseuxi was a more precise thermoregulator and was less sensitive to heat, and P, hasselquistii was the most sensitive to heat exposure, P, guttatus was intermediate, P, puiseuxi maintained relatively stable oxygen consumption and heart frequency in the range of 25-40 degrees C and maintained a lower body temperature at 35 and 40 degrees C, compared with both other species, The nocturnal P, hasselquistii doubled its oxygen consumption between 35 and 40 degrees C and significantly increased its heart and respiration frequencies as a function of ambient temperature, The diurno-nocturnal P, guttatus was intermediate in its responses with both its oxygen consumption and its heart frequency increasing moderately as a function of ambient temperature, The heating/cooling rate ratio is related to the degree of diurnality, I conclude that the physiological responses of the three congeners to increasing ambient temperature are closely related to their distribution and ecology. Biol. 169 1 29-36 AM, embryology, endocrinology, In most male vertebrates, the mullerian ducts, which are the embryonic oviducts, regress completely during embryogenesis or shortly thereafter. Mullerian duct regression is caused by the testicular hormone mullerian inhibiting substance. While the exact mechanism of mullerian duct regression is not well understood, it is theorized to involve active migration of at least some of the ductal epithelial cells following breakdown of the basement membrane, possibly through an interaction between the epithelial cells and the surrounding extracellular matrix. The purpose of the present study was (1) to confirm that mullerian duct regression is characterized by epithelial cell migration and (2) to determine the destination of these cells. The ductal epithelium of male alligator embryos was labelled with the fluorescent cell marker DiI and then allowed to undergo regression in vitro. DiI-labeled mullerian ducts from female embryos served as controls. An in vivo experiment also was performed in which the mullerian ducts of female hatchlings were injected with DiI prior to each female receiving a testicular implant from a male hatchling. Control females received a kidney implant instead. In both the in vitro and the in vivo experiments, DiI-labeled epithelial cells were observed migrating out from the duct during regression. In the in vitro experiment in which regression proceeded more rapidly, epithelial cells entered the mesonephric kidneys and were incorporated into the nephric tubule epithelium. No regression or cell migration was observed in the control mullerian ducts. One of the growth models used (Preece & Baines, 1978) was found to be superior to the classical growth models (exponential, monomolecular, logistic, Gompertz, von Bertalanffy) and gave an adequate fit to all longitudinal measures taken from the embryonic body and embryonic mass. However, measurements taken from the head could not be fitted by growth models but were adequately fitted by weighted least squares cubic splines. Data for the stage of development were best fitted by a sum of 2 exponentials with a transition point. Comparison of the maximum growth rates and parameter values, indicated that the growth data at 30 degrees C could be scaled to 33 degrees C to multiplying the time by a scaling factor of 1.2. This is equivalent to a Q10 of about 1.86 or, after solving the Arrhenius equation, an E++ of 46.9 kJmol-1. This may be interpreted as indicating a common rate-limiting step in development at the 2 temperatures. Head, jaw and throat movements in drinking cycles are described. Postures of neck and head were compared for three water depth (2, 1O and 30 mm). Water expulsion. occurring after the turtle was disturbed, is also kinematically, described. Finally, a drinking mechanism in the turtle is proposed.
