A large, well-circumscribed, hypervascular ovarian mass with multiple fluid-filled vesicles was identified during surgery. A histopathologic diagnosis of ovarian teratoma was made based on the presence of multiple tissues of different embryonic germ-layer origins. The teratoma recurred as a malignant tumor 18 mo later. Res. 24 3 215-226 CCR, anatomy, The multivariate statistical analysis provides a useful method to study neuronal populations. It allows both the objective classification of neuronal types and the study of the morphological variation within a neuronal population. We report a particular example of the use of these techniques on a Golgi study of a complex telencephalic structure, the reptilian anterior dorsal ventricular ridge (aDVR). We present a R mode factor analysis and a cluster analysis on the results of the factor analysis. Sixteen original variables were chosen for the study in order to obtain the greatest information about the cell body, the dendritic field and the location of 96 Golgi stained cells. Six factors were obtained after the R mode factor analysis, the interpretation of which was clear in five of them. This contributed to explain the morphological variation of the neuronal types within the aDVR. The cluster analysis classified the 96 cells into eight groups. Xenoestrogens mimic the natural estrogen 17 beta estradiol and compete for binding to the estrogen receptor. Even though the affinity of o,p' DDT and OP for the estrogen receptor is approximately 1000 fold lower than 17 beta estradiol, the actions of xenoestrogens could be enhanced, if their bioavailability in serum were greater than 17 beta estradiol. To test this hypothesis, the yeast estrogen screen (YES) was created by expressing human estrogen (hER) and two estrogen response elements (ERE) linked to the lacZ gene. The beta galactosidase activity of the YES system was significantly increased after treatment with 17 beta estradiol or the xenostrogens diethylstilbestrol (DES), o,p' DDT, and OP but not with vehicle antiestrogen ICI 164,384, dexamethasone, or testosterone. To determine whether serum proteins affected the bioavailability of natural estrogens compared to xenoestrogens, albumin, sex hormone binding globulin (SHBG), or charcoal stripped serum were added to the YES system and beta galactosidase activity assayed. Albumin and SHBG decreased beta galactosidase activity in the presence of estradiol to a greater extent than DES, o,p' DDT, and OP. Human and alligator charcoal stripped were also effective at selectively reducing beta galactosidase in the presence of xenoestrogens, indicating that serum may serve as a biomarker for sensitivity to xenoestrogens may exert greater estrogenicity than originally predicted. The estrogenic potency of a compound involves its binding affinity, bioavailability in serum, and persistence in the environment. Our data demonstrate the utility of the YES system for identifying and characterizing environmental estrogens. Using high-resolution angioscopy, we have studied, these structures in the beating crocodile heart and correlated their movements with in vivo pressure and flow recordings. The connective tissue outpushings in the pulmonary outflow tract represent an active mechanism used to restrict blood flow into the lungs, thus creating one of the conditions required for a right-to-left shunt, We observed that the foramen of Panizza was obstructed by the medial cusp of the right aortic valve during most of systole, effectively differentiating the left and right aortic blood pressure, During diastole, however, the foramen remained open, allowing pressure equilibration between the two aortas, Contrary to current theories, we found that the left aortic valves were unable to cover the foramen of Panizza during any part of the cardiac cycle, supporting the reversed foramen flow hypothesis, This would ensure a supply of blood to the coronary and cephalic circulation during a complete shutdown of the left side of the heart, such as might occur during prolonged submergence. The highest population of zoosporic fungi was collected from Biomphalaria alexandrina, the lowest from Bulinus truncatus. Copper sulphate was more effective than Bayluscide on the fungal species of both snails. Achlya, Dictyuchus and Saprolegnia were the common genera while Leptolegnia caudata and Pythium ultimum completely missed from treated snails. The total colonies of zoosporic fungi were lowered after. 15 days of treatment. The two molluscicides inhibited the cell division of both tested snails. Here we present a gene sequence showing a high homology to grass pollen allergens of type II/III thought to be expressed in pollen tissue only. Surprisingly, the pollen allergen-like sequence, designated Tri a III, is also expressed in gynoecia of the sexual, male fertile wheat line '(aestivum)-Salmon', whereas the two parthenogenetic and male sterile wheat lines '(caudata)-Salmon' and '(kotschyi)-Salmon' completely lack any Tri a III transcript. Our data suggest a positive correlation between the expression of this clone and the manifestation of male fertility. Northern and in situ hybridization analysis revealed that, in addition to its presence in pollen, Tri a III is expressed in the parenchymatous tissue of '(aestivum)-Salmon' ovaries exclusively at the day of anthesis. This precise temporal and spatial expression pattern suggests a more general function of the pollen allergen-like sequence Tri a III not limited to the exhibition of allergens in pollen grains. Like most Neotropical Oswaldacruzia, Oswaldacruzia venezuelensis sp. n. is characterized by spicules with three principal branches: blade, shoe and fork, and by a division of the fork within the distal third of the spicule length. O. vaucheri Ben Slimane et Durette-Desset, 1993 is the most closely related species due to its caudal bursa of type Il and its cervical alae of the same shape but it differs in the following characters: the position of the papillae of rays 4 situated nearer the papillae of rays 3 rather than rays 5, a higher percentage of the ridges in the oesophageal region, the cervical alae three times longer and sharp and the spicular fork divided less deeply. The cellular basis of regulatory volume decrease (RVD) by mudpuppy (Necturus maculosus) red blood cells (RBCs) was examined. Volume regulation was inhibited by replacing extracellular Na+ with K+. In contrast, addition of gramicidin (5 microM) to the extracellular medium enhanced RVD. The K(+)-channel blocker quinine (1 mM) also inhibited RVD, and this inhibition was reversed by gramicidin (5 microM). In addition, a 0 Ca(2+)-EGTA Ringer blocked RVD, whereas the Ca2+ ionophore A23187 ( microM) enhanced recovery of cell volume. The stretch-activated ion channel antagonist gadolinium (10 microM) inhibited RVD, and this effect was reversed by A23187 (2 microM). Furthermore, the calmodulin inhibitors pimozide (10 microM) and N-(6- aminohexyl)-5-chloro-1-napthalene-sulfonamide (0.1 mM) blocked RVD, and this inhibition was reversed with gramicidin (5 microM). Consistent with these findings, a K(+)-selective membrane conductance was activated by exposing RBCs to a 0.5x Ringer solution (observed with the whole cell patch clamp technique). This conductance was inhibited by quinine (1 mM), gadolinium (10 microM), and pimozide (10 microM). These results indicate that cell swelling activates a K+ conductance by a Ca(2+)-calmodulin-dependent mechanism and that this channel mediates K+ loss during RVD. Unstimulated water and Na+ permeabilities were low along the ventral skin. Hydrosmotic and natriferic responses to oxytocin increased from thorax to pelvis, Unstimulated Na+ conductance was greater in pelvis than in abdomen, the other electrical parameters being essentially similar in both skin fragments. Contribution of shunt conductance to total skin conductance was higher in abdominal than in pelvic skin. Oxytocin-induced increases of total skin conductance, Na+ conductance, and shunt conductance in pelvis were significantly larger than in abdomen, An oscillatory behaviour of the short-circuit current was observed only in oxytocin-treated pelvic skins. Decrease of epithelial thickness and increase of mitochondria-rich cell number were observed from thorax to pelvis, Oxytocin-induced increases of interspaces were more conspicuous in pelvis and abdomen than in thorax. Whole cell patch-clamp recordings and electron micrographs were obtained from cells in Necturus taste buds in lingual slices to study their membrane properties and to correlate these properties with cell ultrastructure. 2. Two different populations of taste receptor cells could be identified: one type possessed voltage-gated Na+ and K+ (noninactivating) currents (group 1 cells); the other type possessed only K+ (inactivating) currents (group 2 cells). 3. The zero-current ("resting") potential (Vo) and whole cell resistance (Ro) of these two types of taste cells differed significantly. For group 1 cells, on average, Vo = -75 mV and Ro = 24.6 G omega, and for group 2 cells, Vo = -49 mV and Ro = 48.9 G omega. The difference in Ro was not explained completely by differences in cell sizes, suggesting that intrinsic membrane properties differed between the populations. 4. Cells injected with biocytin were the electron microscope after tissues were reacted with majority (14 of 16) of cells with voltage-gated Na+ and K+ currents (group 1 cells) were characterized by abundant rough endoplasmic reticulum and dense granular packets in the apical process. These are features of dark cells. All the cells that only possessed K+ currents (group 2 cells) were characterize by well-developed smooth endoplasmic reticulum and an absence granular packets. These features characterize light cells. 5. These findings indicate that there is a good, although not exact, correlation between electrophysiological properties and cell morphotype in Necturus taste bud cells. All dark cells possessed Na+ and K+ currents and thus would be expected to be capable of generating action potentials. Cardiovascular function must be critical to this ability. However, information on cardiovascular function in developing reptiles is lacking. Previous work indicated that in reptiles the effects of temperature on growth and metabolism are largely restricted to early development. This study examined whether the previously observed effects of temperature and different perinatal patterns of metabolism observed in amniotic vertebrates are correlated with cardiovascular function. Embryonic and hatchling carcass mass, heart mass and heart rate (HR) were compared for snapping turtle eggs (Chelydra serpentina) incubated at 24 degrees and 29 degrees C. Incubation time was shorter at 29 degrees C (56.2 days) than at 24 degrees C (71.1 days). Carcass and heart growth showed a sigmoidal pattern at both temperatures. However, cardiac growth showed a relative decrease as incubation proceeded. Incubation temperature significantly affected the HR pattern during development. The HR of embryos incubated at 24 degrees C was constant for most of incubation (51.8 +, - 4.8 min(-1)). A small decrease was observed just prior to and a large decrease immediately following hatching (posthatch, 22.3 +, - 4.1 min(-1)). At 29 degrees C embryonic HR was greater than at 24 degrees C early in development (72.3 +, - 3 min(-1)). The HR steadily decreased to values equivalent to those at 24 degrees C. The HRs of 24 degrees C and 29 degrees C hatchlings were not different. Cardiac output (estimated as the product of heart mass and HR) increased rapidly during early development and then slowed dramatically at both temperatures. These data are consistent with the suggestion that temperature exerts its effects primarily early in development. Furthermore, the changes in cardiovascular function are correlated with metabolic changes in hatching vertebrates. Incubation time increased with increasing egg mass (allometric exponent = 0.14, r(2) = 0.31). However, significant heterogeneity was found within the Reptilia for this character. If periods of developmental arrest are excluded the orders Crocodylia, Testudinata, and the suborder Serpentes have the shortest incubation times and the lizards the longest. Significant differences within lizard families were also observed (Iguanidae, Agamidae, Gekkonidae, and Lacertidae have shorter incubation times than the Varanidae and Teiidae). Preliminary analyses indicate the heterogeneity in incubation time observed within the Squamata is correlated with differences in brain mass; this is similar to findings for mammals. The variation in incubation time indicates the existence of differences in growth rates in reptilian groups and suggests a partial explanation for the observed distribution of viviparity within the Squamata. Capture-recapture procedures were carried out during 27 periods over 3 years in a large population of female asp vipers, Vipera aspis (317 marked individuals). Catchability was greater in reproductive females (2.64 +/- 1.74 (SD) captures/year) than in nonreproductive females (1.76 +/- 1.02). Three different methods were used to estimate breeding frequency: (i) the proportion of reproductive to nonreproductive females for each period (noncorrected estimates, NC) was measured, (ii) the ratio of the number of reproductive females to the total number of females was estimated using the CAPTURE program (catchability-corrected estimates, CE); (iii) breeding frequencies of marked individuals were observed in the field for 3 years (observed breeding frequency, OF). The results obtained with the three methods were different. The NC procedure indicated that 63% of females breed each year, suggesting an annual-biennial cycle. The CE (34% reproductive females) and OF (34% reproductive females) procedures both suggested a triennial cycle. The OF method was the most accurate; breeding frequencies were overestimated with the NC method. The NC method is the one most commonly used, and estimates of breeding frequency in snakes obtained in these studies may be imprecise. We conclude that differential catchability between reproductive and nonreproductive females should be taken into account in estimating breeding frequencies. The sutures in most caudal vertebrae are fully closed at hatching, but closure of remaining sutures occurs later in ontogeny. Closure of cervical sutures is a consistent indicator of morphological maturity in Alligator mississippiensis, Alligator sinensis, Osteolaemus tetraspis, and Crocodylus acutus; the final transformation is the closure of the axial neurocentral suture, which occurs after the closure of the axis odontoid suture. Because these transformations occur near the end of ontogeny in all three taxa, regardless of maximum size, closure of these sutures is a size independant criterion of maturity; however, it is not certain if suture closure indicates the stoppage of growth. These transformations are readily identifiable in fossils, permitting the objective characterization of maturity in fossil crocodylians and possibly at least some of their closer extinct relatives. In this genus, alpha 2-M constitutes 11-13% of total plasma protein (approximately 2.2-3.9 mg/mL). Analysis of blood samples collected at approximately monthly intervals from individual tuatara indicated that the plasma level of alpha 2-M remains fairly constant. The subunits of tuatara alpha 2-M have an apparent molecular mass of approximately 160 kDa as determined by SDS-polyacrylamide gel electrophoresis and the intact protein is an oligomer that contains inter-chain disulfide bonds. N-terminal sequence analyses of tuatara alpha 2-M revealed a distinct similarity to alpha-macroglobulins of other vertebrates and that at least two types of alpha 2-M subunits are present in plasma of tuatara. The Directorate of Animal Health is responsible for disease control measures in respect of Salmonidae, while the Directorate of Plant and Quality Control regulates the introduction of exotic and unwanted aquatic species into the RSA. The activities and interests of commercial aquaculture are coordinated through the South African Aquaculture Organization and a national coordinating committee under the control of the national Department of Agriculture. Four diseases of trout are notifiable under the terms of the Animal Diseases Act (Act 35 of 1984), namely viral haemorrhagic septicaemia, infectious pancreatic necrosis, infectious haematopoietic necrosis and bacterial kidney disease. The zoosanitary measures in respect of these diseases require State intervention after detection of occurrence. Sampling procedures for the diagnosis of diseases, maintenance of the disease-free status of trout farms accredited for export purposes and requirements for the import of ova are based on the standards recommended in the International Aquatic Animal Health Code of the Office International des Epizooties. Zoosanitary control of other aquatic species not related to aquaculture--especially ornamental fish--may be unnecessary. An embargo, through existing legislation on the importation of unwanted species, has been the only means of protecting inland species. The author concludes by evaluating the need for, and the feasibility of, additional legislation for the zoosanitary control of other aquatic species. The objectives of this study were to clone the POSP cDNA, obtain the full-length cDNA and protein sequence, examine tissue specificity and species distribution, characterize its regulation, and establish its identity by comparison to other known protein, RNA, or DNA sequences. A full-length cDNA of 2022 base pairs was obtained with an open reading frame of 1581 nucleotides, coding for a deduced protein of 527 amino acids (57 970 M(r)). The deduced protein contained three potential N-glycosylation sites, a consensus heparin-binding site, and potential O-glycosylation sites. Amino acid analysis of POSP-E3 confirmed the presence of a 21-amino acid signal sequence. Northern blot analysis revealed an oviduct-specific mRNA species of 2.25 kb in the infundibulum (INF), ampulla (A), and isthmus (I). An mRNA of similar size was detected in the oviduct of the sheep, cow, and rabbit, and one of slightly greater size (2.8 kb) in the mouse and hamster oviduct but not in the horse or alligator oviduct. Dot blot analysis indicated that steady-state levels of POSP mRNA were significantly greater (p = 0.0001) in the A than in the INF or I regardless of day of the estrous cycle and were greater on Day 0 (estrus; p = 0.0001) regardless of location. Further, steady-state mRNA levels were significantly increased (p = 0.02) on Days 0 and 1, declining rapidly to Day 2 through Day 15 of the estrous cycle. Steady-state POSP mRNA levels were significantly greater (p < 0.003) in ovariectomized gilts treated with estradiol valerate than those treated with other steroid regimens, vehicle, or no treatment (Control), consistent with estrogen control of mRNA expression. The POSP protein exhibited significant identity to oviductal glycoproteins from the baboon, cow, hamster, human, mouse, and sheep, to several mammalian nonoviductal glycoproteins; and to several chitinases. POSP joins a growing subfamily of the chitinase gene family that lacks chitinase enzymatic activity. Oochoristica macallisteri sp. n. belongs to a group of species of Oochoristica possessing fewer than 25 testes in a single cluster and circular suckers, namely, O. elongata, O. junkea, O. lygosomae, O. lygosomatis, O. novaezealandae, O. parvogenitalis and O. sobolevi. The new species can be readily differentiated from the other species of this group by number of lobules of the ovary or number of proglottids and from other species from California by number of testes or shape of vitellarium. (Nematoda: Pharyngodonidae), a new oxyurid nematode, discovered in the large intestine of the gecko Gehyra oceanica is described and illustrated. Nine of 11 adult female specimens of G. oceanica collected on Guam, Mariana Islands, Micronesia, harbored a total of 480 specimens of S. gehyrae; prevalence of infection was 82% (mean intensity 53, range 6-179). Spauligodon gehyrae represents the 33rd species to be described for this genus and is distinguished by the presence of truncated egg ends, the short stiff tail spike of the female, and the presence of a spicule in the male. This is the second species of Spauligodon to be described from Oceania. Spauligodon hemidactylus n. sp. represents the 31st species to be described for this genus and is distinguished by the low number of tail filament spines of the female and the presence of unequal knobs on the egg. This is the first report of the genus Spauligodon from Oceania.
