Procedure Six green sea turtles (Chelonia mydas) spontaneously infected with cardiovascular flukes (Digenea: Spirorchidae) were treated orally with praziquantel, and necropsied 3 or 7 days later to look for flukes in the heart and major blood vessels, Six similar animals were maintained as untreated controls. Results Absence of flukes in treated, but not control turtles, indicated that a one day course of treatment at a dose rate of 3 x 50 mg, kg body weight is effective. Conclusion: This result should be of value for preventing disease in wild caught green turtles introduced into farms or aquaria. W Adnyana, James Cook Univ N Queensland, Australian Inst Trop Vet & Anim Sci, Townsville, Qld 4811, Australia Journal Article Adnyana, W.;Ladds, P.W.;Blair, D. 1997 Observations of fibropapillomatosis in green turtles (Chelonia mydas) in Indonesia Australian Veterinary Journal 75 10 737-742 Reptilia, testudines, green turtle, Chelonia mydas, infection, tumour, pathology, fibropapilloma Objective To determine the prevalence and manifestations of fibropapillomatosis in green turtles in Indonesia, to identify any relationship between fibropapillomatosis and concurrent parasitic infection, to ascertain the effect of fibropapillomatosis on health, and to examine whether environment might have an effect on the prevalence of fibropapillomatosis. Procedure 4407 green turtles (Chelonia mydas) and 401 hawksbill turtles (Eretmochelys imbricata) were examined. The occurrence of fibropapillomatosis was correlated with sex, maturity, curved carapace length, body weight curved carapace length ratio, the number and distribution of tumours on the skin, parasite burdens, some haematological variables and the region of capture. Results Fibropapillomatosis was seen only in green turtles, and the overall prevalence in these was 21.5%. This prevalence increased with the curved carapace length up to 85 cm. The average number of tumours per affected turtle was 5 + - SD 4.1 (range, 1 to 29), and was negatively correlated with the body weight curved carapace length ratio (rs = -0.8; P = 0.001). The red blood cell count in turtles with fibropapilloma was lower than in non-fibropapilloma turtles captured and examined at the same time (P = 0.001). The prevalence of fibropapilloma in turtles captured near densely populated, industrial regions (26.3%) was greater than in turtles from sparsely populated areas (17.7%). Conclusion Fibropapillomatosis in green sea turtles in Indonesia is of moderate occurrence; young mature turtles (curved carapace length = 85 cm) are most frequently affected. Fibropapilloma adversely affects health of turtles. Fluke infestation seems not to be a causal factor, but viral infection, perhaps with concurrent stress of environmental origin, seems likely. W. 1997 Genetic variation in some populations of the golden-striped salamander, Chioglossa lusitanica (Amphibia: Urodela), in Portugal Biochem Genet 35 11-12 371-81 Genetic variation in the golden-striped salamander (Chioglossa lusitanica) was assessed in 231 individuals from four Portuguese populations by means of horizontal starch gel electrophoresis and isoelectric focusing. Three of 19 enzyme systems, representing 21 presumptive loci, were found to be polymorphic: phosphoglucomutase 1 (PGM1), peptidase B (PEPB), and peptidase D (PEPD). The observed average heterozygosity in Chioglossa lusitanica (0.027) is significantly lower than that observed for other amphibians, either urodeles or salamandrids. Differences in allele frequencies and the presence of private alleles are indicative of a high degree of population differentiation. To investigate this phenomenon, the estrogen receptor (ER) from the American alligator (aER) or human (hER) was incubated with 17 beta-estradiol in the presence of selected environmental chemicals individually or in combination. The environmental chemicals included the insecticide chlordane, which has no estrogenic activity. and the pesticides dieldrin and toxaphene, which have very weak estrogenic activity. Chlordane, dieldrin, and toxaphene individually demonstrated no appreciable displacement of 17 beta-estradiol from aER and hER at the concentration tested. A combination of these chemicals inhibited the binding of 17 beta-estradiol by 20 to 40%. Alachlor, a chemical recently discovered to have weak estrogenic activity, also displaced 17 beta-estradiol more effectively in combination with dieldrin than alone. These results indicate that combinations of some environmental chemicals inhibit 17 beta-estradiol binding in a synergistic manner. This suggests that the ER may contain more than one site for binding environmental chemicals. The possibility that the ER binds multiple environmental chemicals adds another level of complexity to the interaction between the environment and the endocrine system. Journal Article Arntzen, J. Over a range of myocardial power outputs at 5 and 15 degrees C we find that turtle hearts perfused with anoxic saline do not show a particularly outstanding ability to produce ATP anaerobically. Furthermore, at 15 degrees C anoxia reduced the ATP turnover rate to 50% of the normoxic rate. At 5 degrees C the anoxia-induced depression of ATP turnover was even more pronounced, being 4-fold lower than the normoxic rate. In addition, anoxia at 5 degrees C reduced the basal metabolic rate of the turtle heart. We conclude that long-term cardiac tolerance of hypoxia in this species is more likely related to metabolic depression rather than to an exceptional anaerobic performance. V. 1997 Motor patterns and kinematics during backward walking in the pacific giant salamander: evidence for novel motor output J Neurophysiol 78 6 3047-60 Kinematic and motor patterns during forward and backward walking in the salamander Dicamptodon tenebrosus were compared to determine whether the differences seen in mammals also apply to a lower vertebrate with sprawling posture and to measure the flexibility of motor output by tetrapod central pattern generators. During treadmill locomotion, electromyograms (EMGs) were recorded from hindlimb muscles of Dicamptodon while simultaneous high-speed video records documented movement of the body, thigh, and crus and allowed EMGs to be synchronized to limb movements. In forward locomotion, the trunk was lifted above the treadmill surface. The pelvic girdle and trunk underwent smooth side-to-side oscillations throughout the stride. At the beginning of the stance phase, the femur was protracted and the knee joint extended. The knee joint initially flexed in early stance and then extended as the foot pushed off in late stance, reaching maximum extension just before foot lift-off. The femur retracted steadily throughout the stance. In the swing phase, the femur rapidly protracted, and the leg was brought forward in an "overhand crawl" motion. In backward walking, the body frequently remained in contact with the treadmill surface. The pelvic girdle, trunk, and femur remained relatively still during stance phase, and most motion occurred at the knee joint. The knee joint extended throughout most of stance, as the body moved back, away from the stationary foot. The knee flexed during swing. Four of five angles showed significantly smaller ranges in backward than in forward walking. EMGs of forward walking showed that ventral muscles were coactive, beginning activity just before foot touchdown and ceasing during the middle of stance phase. Dorsal muscles were active primarily during swing. Backward locomotion showed a different pattern; all muscles except one showed primary activity during the swing phase. This pattern of muscle synergy in backward walking never was seen in forward locomotion. Also, several muscles demonstrated lower burst rectified integrated areas (RIA) or durations during backward locomotion. Multivariate statistical analysis of EMG onset and RIA completely separated forward and backward walking along the first principal component, based on higher RIAs, longer durations of muscle activity, and greater synergy between ventral muscles during early stance in forward walking. Backward walking in Dicamptodon uses a novel motor pattern not seen during forward walking in salamanders or during any other locomotor activity in previously studied tetrapods. The central neuronal mechanisms mediating locomotion in this primitive tetrapod are thus capable of considerable plasticity. We identified 56 compounds in V-SDE extracts and 13 in DHS extracts. Volatile profiles were similar for all alligator meat samples regardless of alligator size (length). However, concentration of volatiles increased as alligator size increased. Most volatile compounds were lipid-derived aldehydes, which made up 73.9-79.4% of total volatiles in V- SDE extracts and 82.4-89.2% in DHS extracts. Hexanal, the most abundant compound, and (Z)-4-heptenal were considered responsible for the green, grassy and fishy, rancid off-odor of cooked alligator meat samples. J. 1997 Antibodies against some viruses of domestic animals in southern African wild animals [In Process Citation] Onderstepoort J Vet Res 64 2 95-110 Twenty-four species of South African wild animals were tested for the presence of antibodies against the viruses of 16 common diseases of domestic animals. Positive results were obtained for African horsesickness, equine encephalosis, equid herpes virus-1, infectious bovine rhinotracheitis, Allerton disease (Herpes mammillitis), lumpy skin disease, parainfluenza, encephalomyocarditis, bluetongue, Wesselsbron disease, bovine ephemeral fever, and Akabane disease complex. No antibodies could be demonstrated against the viruses of equine influenza, equine infectious anaemia, equine viral arteritis and Rift Valley fever. The negative results substantiate observations that the latter diseases, with the exception of equine viral arteritis, are absent in South Africa. The number of animal species found positive for a specific virus, ranged from 0-16. No antibodies were found in crocodiles and warthogs, whereas antibodies against Wesselsbron and bovid herpes virus-1 were present in 16 species. Antibodies against viruses of horses were found almost exclusively in zebras and, although elephants reacted to African horsesickness, no neutralizing antibodies against it could be demonstrated in their sera. Zebras were also found to be positive for Wesselsbron and Akabane, which are usually regarded as viruses of ruminants. Antibodies against most viruses were encountered in all vegetation zones in South Africa but, as a rule, most viruses were more prevalent in the high-rainfall zone in KwaZulu- Natal. The nasal cavity was largely replaced by granulomatous inflammatory tissue, and most internal organs had nodular granulomas. Typical mother and daughter sphaerules of M. amphibiorum occurred in these nodules which were due to granulomatous inflammation as well as areas of more active mixed inflammation with necrosis. Tissue homogenate from the spleen was inoculated into two cane toads (Bufo marinus), and one toad became infected with M. amphibiorum. The patient developed swelling and ecchymoses in the affected extremity, and cutaneous necrosis of the envenomated fingertip. There was no significant coagulopathy. He received 20 vials of specific antivenin (Schlangengift-Immunserum Behring Zentralafrika, Behringwerke, Marburg, Germany) and debridement of devitalized finger tissue. The only permanent sequelae were cutaneous scarring and permanent loss of the fingernail on the envenomated finger. Exotic snakebite is a rare presenting problem in emergency departments. The initial approach to a patient envenomated by an exotic venomous snake is discussed. Use of antivenin and supportive care are emphasized. In comparison with other countries there is a normal population density in the three places. Caiman crocodilus fuscus was found in both rivers. Crocodylus acutus occurs in the three zones. The isolation of hemoglobin mRNA was based on degenerate primers' PCR in combination with 5'- and 3'-RACE protocol. The full length cDNA is 615 bp with the ATG start codon at position 53 and TGA stop codon at position 495; The AATAAA polyadenylation signal is found at position 599. The deduced polypeptyde contains 146 amino-acid residues. The predicted amino acid sequence shares 83% identity with the beta-globin of a related specie, the aquatic turtle C. p. belli. Otherwise, identity is higher when compared with chicken beta-Hb (80%) than with other reptilian orders (Squamata, 69%, and Crocodilia, 61%). Compared with human HbA, there is 67% identity, and at least three amino acid substitutions could be of some functional significance (Glu43 beta-->Ser, His116 beta-->Thr and His143 beta-->Leu). To our knowledge this represents the first cDNA sequence of a reptile globin gene described. A fishing hook was lodged in the corner of its mouth, monofilament line trailed from its cloaca. and radiography revealed that the turtle had ingested two additional hooks and a large sinker. The hemogram showed leukocytosis. At exploratory celiotomy, the fishing line was seen to have acted as a linear foreign body and had perforated the intestines. Multiple enterotomies were performed to remove the sinker and line, and perforations were repaired. Two of the hooks could not be surgically or endoscopically retrieved. Blood lead concentration was 3.6 ppm prior to the start of chelation therapy with calcium disodium ethylenediaminetetraacetic acid and declined to undetectable levels within 6 wk. HCO3 transporters are important for regulation of intracellular pH (pHi) in most cells and also thereby regulate blood pH. This electrogenic Na/HCO3 cotransporter was first discovered using perfused Ambystoma tigrinum (salamander) renal, proximal tubules. This novel cotransporter mediates the movement of one Na+ ion with several HCO3- ions, making it electrogenic, is blocked by stilbene compounds, but does not depend on intra- or extracellular Cl-. This and similar cotransporters have been found in a number of tissues and cell types. Recently, we used Xenopus-laevis oocytes to expression clone the salamander renal electrogenic Na Bicarbonate Cotransporter (NBC). Using microelectrodes to monitor membrane potential (Vm) and intracellular pH (pHi), we followed oocyte expression after injecting poly (A)+, fractioned poly (A)+, or cRNA. All experimental solutions contained 100 microM ouabain to block the Na+/K+ pump. Our expression assay was to apply 1.5% CO2/10 mM HCO3- (pH 7.5), allow pHi to stabilize from the CO2-induced acidification, and then remove bath Na+. Removing bath Na+ from native oocytes and water-injected controls, hyperpolarized the oocytes by approximately 5 mV and had no effect on pHi. However, for oocytes injected with poly (A)+ RNA, removing Na+ transiently depolarized the cell by approximately 10 mV and caused pHi to decrease; both effects were blocked by 4,4'-diisothiocyano-2,2'- stilbenedisulfonate (DIDS) and required HCO3-. Electrophoretic fractionation of the poly (A)+ RNA, enriched the expression signal. From the optimal expression-fraction, we constructed a size-selected cDNA library in pSPORT1. Screening our Ambystoma library yielded a single clone (aNBC). We could detect expression 3 days after injection of NBC cRNA. In aNBC-expressing oocytes, adding CO2/HCO3-elicited a large (> 50mV) and rapid hyperpolarization, followed by a partial relaxation as pHi stabilized. Na+ removal in CO2/HCO3-depolarized the cell by > 40mV and decreased pHi, aNBC encodes a protein of 1035 amino acids with several putative membrane-spanning domains, and has a low level of amino-acid homology (approximately 30% to the AE family of Cl- HCO3 exchangers. aNBC is the first member of a new family of Na(+)- linked HCO3- transporters and, together with the AE family, defines a new superfamily of HCO3- transporters. Using aNBC to screen a rat- kidney cDNA library, we identified a full-length cDNA clone (rNBC), rNBC encodes a protein of 1035 amino acids, is 86% identical to aNBC, and can be functionally expressed in oocytes. When morphology was analyzed alone, Gavialis was the sister taxon of all other extant crocodylians whether or not fossil ingroup taxa were included, and a sister-group relationship between Gavialis and Tomistoma was significantly less parsimonious. In combination with published sequence and restriction site fragment data, Gavialis was the sister taxon of all other living crocodylians, but the position of Tomistoma depended on the inclusion of fossil ingroup taxa; with or without fossils, preferred morphological and molecular topologies were not significantly different. Fossils closer to Gavialis than to Tomistoma can be recognized in the Late Cretaceous, and fossil relatives of Tomisioma are known from the basal Eocene, strongly indicating a divergence long before the Late Tertiary. Comparison of minimum divergence time from the fossil record with different measures of molecular distance indicates evolutionary rate heterogeneity within Crocodylia. Fossils strongly contradict a post-Oligocene divergence between Gavialis and any other living crocodylian, but the phylogenetic placement of Gavialis is best viewed as unresolved. Leidyosuchus riggsi is a nomen dubium, L. gilmorei is a junior synonym of L. canadensis, and Diplocynodon stuckeri is a junior synonym of L. wilsoni. Phylogenetic analysis of 164 morphological characters indicates a basal position within Alligatoroidea for L. canadensis. The name Listrognathosuchus is established for L. multidentatus, which is an alligtoroid, but difficult to characterize further because of its incompleteness. The remaining taxa form a clade herein named Borealosuchus; this group is related to, but not within, a monophyletic group that includes Alligatoroidea and Crocodyloidea. A close relationship between these taxa and Diplocynodon is rejected. The amino acid composition of tuatara Vg is similar to that of other vertebtate Vgs and contains a large proportion of serine (13.7 mol/100 mol of total amino acid). The amino acid sequences of the N-terminus of mature Vg (33 residues) and of several trypsin- and CNBr-generated peptides were determined. Six peptide sequences obtained from tuatara Vg could be aligned with Vg sequences from other vertebrates. Reduced and non-reduced forms of tuatara Vg have the same apparent molecular mass (approximately 218 kDa) when resolved by SDS-PAGE, indicating that inter-chain disulfide bonds are not a feature of the molecule in this species. Western blot analysis with anti-tuatara Vg antiserum indicated that at least some epitopes are shared among Vgs of turtle, alligator and tuatara. We also examined sperm storage in the reproductive tracts of both sexes. Males in south-eastern Queensland had a testicular cycle that was strongly seasonal, with testicular regression during the wet summer. Males from Papua New Guinea had a continuous testicular cycle, in which sperm was present in some animals every month. Females from south-eastern Queensland had no stored sperm or sperm storage receptacles in their oviducts. Sperm was found in only one of 28 female reproductive tracts. The sperm was located in the lumen, suggesting recent insemination. In contrast, the presence of sperm in the ductus deferens of most males in both populations indicated that males store sperm. This facilitates the apparent dissociation between the male and female reproductive cycles that has been reported previously in south-eastern Queensland populations. Male B. irregularis must store sperm for at least six months, from the time of maximal testicular development in autumn, until ovulation occurs in spring and early summer. This pattern of reproduction in B. irregularis fits neither a postnuptial nor a prenuptial pattern, as has been observed in a few other tropical snakes. Oochoristica jonnesi sp. n. has few characteristics in common with African species of Oochoristica; rather, it belongs to that group of species possessing fewer than 25 testes in a single cluster and circular suckers, namely, O. junkea, O. lygosomae, O. lygosomatis, O. novaezelandae, and O. sobolevi. The new species can be readily differentiated from the other species of this group by number of lobules of the ovary or number of osmoregulatory canals. This hypothesis, based on a developmental model of the organization of the tetrapod limb, is in accordance with the fossil evidence, and may supersede traditional Haeckelian views based on recapitulatory paradigms. The homologous relationships of carpal elements reveal the existence of two carpal patterns-one plesiomorphic and one apomorphic-in the crocodilian lineage. Phylogenetic change is explained causally by alterations of the osteogenesis of the distal carpals 2 and 3, which remain unossified in extant crocodile adults. This implies that crocodilian limb evolution is constrained by a process of paedomorphosis. This modification of the architecture of the crocodilian hand is a terminal event of its evolutionary history, affecting only eusuchian crocodiles. The results of this study contest the traditional view that the skeletal pattern of the crocodilian limb has been conserved unchanged since the Triassic. R. 1997 Epidemiological and clinical aspects of snakebite in Belo Horizonte, southeast Brazil Rev Inst Med Trop Sao Paulo 39 2 113-8 Epidemiologic and clinical aspects of 310 hospitalized snakebite patients and 310 matched controls were described, over a seven years period, from an emergency hospital in Belo Horizontal, Southeast Brazil. The diagnosis was based upon clinical picture or actual snake identification. Fifty six percent of victims were bitten by the snakes of genus Bothrops, 32.0% by Crotalus, 1.0% by Lachesis and 10.0% undetermined. During the study period, stable number of cases and marked seasonal variation were noted. In comparing cases of snakebite and controls, those from a rural area or who were involved in agricultural labor activity were identified as a high risk group, with an odds ratio (OR) of 14.7 and 6.7, respectively, in favor of being bitten. Upon treatment, snakebite patients were 13.5 times more likely to have had early anaphylactic reactions than their controls, with a higher association in the age group > or = 20 years (OR = 30.3). Increased risks were also detected for pyrexia (OR = 11.7), with a marked association in the group under 19 years old (OR = 16.6). Severe cases of snakebite are an important treatable cause of morbidity in Brazil but therapy may be potentially life threatening. The higher case-fatality ratio encountered, compared to national statistics may be due the representativeness of the more severe cases who sought hospitalization. Preventing snakebite and early referral of those who are bitten is proposed.
