Shell morphogenesis begins at embryonic stage 15 and the shape of the shell is mostly completed by embryonic stage 19. The carapace anlagen arises as a thickening of the skin in the dorsal part of the mid-trunk region between the anterior and posterior limbs. This thickening extends ventro-laterally to form ridges at the margins of the carapace. Each ridge forms as a thick epidermal placode over a condensation of mesenchymal cells. The epidermis behind the advancing margins of the carapace is cuboidal or columnar but does not form placodes. The margins of the carapace expand rapidly in all directions. The plastron anlagen is derived from epidermal cells localised in the latero-ventral regions between the fore- and hind-limbs. Plastron placodes are present laterally, while the mid-ventral and central epidermis remains cuboidal or columnar but does not form placodes at embryonic stage 16. The plastron thickening rapidly moves from a latero-ventral position to a flat ventral position between embryonic stages 16 and 19. Dermal-epidermal anchoring complexes occur throughout placodes of both the carapace and plastron, but are rare in non-placode areas. The accumulation of a dense mesenchyme beneath the shell epidermis forms a dermal cushion that surrounds the body cavity. The superficial dermis close to the epidermis is made of mesenchymal fibroblasts at embryonic stage 19, although the inner-most areas contain bipolar fibroblasts and extracellular fibrils. Scutes with set-rations at their borders form as invaginations of the epidermis into the dermis in the mid-dorsal areas of the embryo at embryonic stages 18-19. Dermal-epidermal anchoring complexes are located around the infoldings that form the scutes of the hinge region. The epidermis of the shell has 2-3 suprabasal cells at embryonic stages 19-22, and lacks keratinisation before embryonic stage 22 when it has 4-6 suprabasal layers with 2-3 external layers made of flat cells. The dermis thickens and has numerous collagen fibrils after embryonic stage 19. The formation of dermal bones begins at embryonic stage 18-19 in the plastron. Only small areas of the carapace near to the bridge have begun to form dermal bone at embryonic stage 19. Animals-10 healthy snakes. Procedure-Various stains, including Wright-Giemsa, benzidine peroxidase, Sudan black B, chloroacetate esterase, cc-naphthyl butyrate esterase, acid phosphatase, leukocyte alkaline phosphatase, periodic acid-Schiff with diastase, and toluidine blue, were used to stain leukocytes differentially on multiple blood smears. Electron microscopy also was performed. Results-Lymphocytes were the most commonly observed leukocyte and could be distinguished from thrombocytes, using periodic acid-Schiff stain with diastase. Azurophils also were commonly observed; their granules stained with peroxidase. Eosinophils were not identified; however, 2 morphologic variations of heterophils were seen in the blood of ail snakes and were considered the same cell type at different stages of cytoplasmic granule development. Heterophil granules were better preserved, using a one-step Wright-Giemsa method that did not require alcohol fixation prior to staining. Degranulated heterophils were observed in all preparations. Conclusions-Most leukocytes of eastern diamondback rattlesnakes can be identified easily on Wright-Giemsa-stained preparations. However, hematologic stains that do not require alcohol fixing prior to staining may be preferred for leukocyte evaluation in certain reptiles. A limited degree of heterophil maturation may continue in the blood of healthy snakes. This, along with degranulation of heterophils, may result in a Variable staining pattern in this cell type, regardless of the stain used. Clinical Relevance-Results provide baseline data for use in hematologic testing in diagnosis of disease and monitoring of treatment of sick or injured snakes. Most of the cases occurred between April and September (86.2 per cent), with peaks in May (25.7 per cent) and July (20.2 per cent), and very few between November and February (3.6 per cent). Forty-two per cent of the dogs were poisoned in the evening (18.00 to 22.00), with a relative risk of 6.85, 17.4 per cent between 22.00 and 02.00 and 16.5 per cent between 14.00 and 18.00. The median age of the dogs was three years, and almost 80 per cent of them were from rural households. German shepherd dogs and rottweilers were over-represented (relative risk 1-98 and 1.87 respectively), and mongrel dogs and pinschers were under-represented (relative risk 0.41 and 0.53 respectively). Fifty-six per cent of the bites were on the head (excluding the mouth, lips and pinnae), 16.5 per cent an the front limbs, 9.7 per cent on the mouth and lips, 8 per cent on the hindlimbs, 4.4 per cent were submandibular and 5.4 per cent were at other sites. The main clinical signs were local swelling (98.2 per cent) and oedema (94.5 per cent), panting (45.7 per cent), tachypnoea (42.5 per cent), pain (34.9 per cent), tachycardia (29.8 per cent), lameness (25.7 per cent), and lymphadenomegaly (23.9 per cent). The mortality rate was 3.7 per cent. The most common haematological abnormalities were neutrophilia (67.6 per cent), leucocytosis (54.9 per cent), thrombocytopenia (51.9 per cent), increased haematocrit (47.6 per cent), and a left shift of neutrophils (37.8 per cent). Many biochemical abnormalities were observed, of which the most common were high activities of lactate dehydrogenase (84.6 per cent), creatine kinase (69 per cent), gamma-glutamyltransferase (40 per cent) and aspartate aminotransferase and high concentrations of globulin. phosphate and total bilirubin (33.3 per cent in each case). This is a new host and locality record for A. Bufonis which has only previously been recorded from amphibians in the Orient. Possible mechanisms for the introduction of A. Bufonis to Hawaii, and its transmission to the toad, are discussed. Almost 98 % of toads were infected with a mean intensify of: 28.6 acanthocephalans per infected toed. There was a significant negative correlation between host length and intensity of infection with subadult toads having significantly higher infection levels than adult male and female loads. We measured the effects of hemolysis on nine plasma analytes in 10 clinically normal common green iguanas (Iguana iguana). Blood samples with moderate and marked hemolysis were produced from each iguana by freezing, centrifuging, and decanting plasma from a portion of each blood sample, and combining the nonhemolyzed plasma with different amounts of hemolyzed plasma from the same individual. Moderate hemolysis significantly increased plasma phosphorus levels. Marked hemolysis significantly increased plasma values of potassium, phosphorus, total protein, and aspartate aminotransferase. The severity of hemolysis must be considered when interpreting values for these analytes in iguana plasma. This organism also is suspected, based on electron microscopic findings, to be the cause of mortality of larval tiger salamanders in a pond over 200 km to the north during the same year. Salamanders developed a generalized viremia which resulted in various lesions including: necrotizing, vesicular and ulcerative dermatitis; gastrointestinal ulceration; and necrosis of hepatic, splenic, renal, lymphoid, and hematopoietic tissues. In cells associated with these lesions, large lightly basophilic cytoplasmic inclusions and vacuolated nuclei with marginated chromatin were consistently found. Virus was isolated from tissue homogenates of infected salamanders following inoculation of epithelioma papilloma cyprini (EPC) cells. The virus, provisionally designated Regina ranavirus (RRV), was initially identified as an iridovirus by electron microscopy. Subsequent molecular characterization, including partial sequence analysis of the major capsid protein (MCP) gene, confirmed this assignment and established that RRV was a ranavirus distinct from frog virus 3 (FV3) and other members of the genus Ranavirus. Intraperitoneal inoculation of 5 x 10(6.23) TCID50 of the field isolate caused mortality in inoculated salamanders at 13 days post infection. Field, clinical, and molecular studies jointly suggest that the etiological agent of recent salamander mortalities is a highly infectious novel ranavirus. One hundred twenty-six Pharyngodon oceanicus sp. n. were found in the large intestines of 14 of 37 adult Gehyra oceanica collected from the Cook and Society islands. Pharynagodon oceanicus sp. n. represents the thirty-first species of the genus and the second species to be described from islands of the Pacific Ocean. It can be distinguished from all other species of Pharyngodon by the shape of the egg and by the presence of a cluster of spines at the base of the tail of the female. Two snakes had tumours of two or three different types, but the other 34 snakes had only one type. All affected animals were adults and their average time on exhibit at the NZP was 108.9 months, Malignant neoplasms (79.5%) outnumbered benign neoplasms (20.5%). Of the malignant: tumours, 19 (61.3%) were considered to have arisen in mesenchymal tissues, 11 (35.5%) were of epithelial origin, and one (3.2%) was derived from neuroectodermal tissues. All the benign neoplasms were of epithelial origin. Neoplasms of the lymphoid and haematopoietic tissues were the most common (12 cases), followed by tumours of the liver and biliary tract (seven cases) and the gastrointestinal tract (four cases). Complete recovery of function after two years Zoo Biology 18 2 141-146 Reptilia, serpentes, northern Pacific rattlesnake, Crotalus viridis oreganus, predation, strike, searching Six northern Pacific rattlesnakes (Crotalus viridis oreganus) rescued from substandard husbandry conditions were shipped from Woodland Park Zoological Gardens to the University of Colorado in December 1994. The snakes accepted prey and rapidly gained weight, but their searching behavior after predatory strikes was seriously depressed. We administered repeated tests of strike-induced chemosensory searching (SICS) finding that the snakes improved steadily with each succeeding test, until performance eventually reached the level characteristic of this taxon. Although the snakes reached normal body weight within 4 months of exposure to proper husbandry conditions, normal SICS was not seen until 2 years had elapsed. Release of these snakes into natural habitat prior to recovery of SICS may well have resulted in the demise of the animals, suggesting that rehabilitation efforts must consider recovery of essential behavior patterns that can take far longer than recovery of body weight. Physical abnormalities included weight loss, sunken eyes, and muscle atrophy. Biochemical abnormalities determined from blood sampling included marked azotemia and hyperosmolality, which were attributed largely to accumulation and retention of nitrogenous wastes. Moderate hypercholesterolemia, hypophosphatemia, and increased aspartate transaminase activity, and mild hyperchloremia, hypocalcemia, hyperbilirubinemia and anemia also were observed, compared with results obtained from other tortoises sampled at the same time. The lack of, or only mild, alterations in most laboratory data exemplified the high degree of physiological adaptation tortoises can undergo when deprived of food and water for a prolonged period. Glucose undergoes net reabsorption involving entry across the luminal membrane by a Na+-dependent, carrier mediated process. However, entry may occur down an electrochemical gradient and exit at the basolateral membrane, against an electrochemical gradient. Amino acids may undergo both net reabsorption and net secretion. Taurine reabsorption involves entry across the luminal membrane against an electrochemical gradient via two independent electrogenic, Na+ and Cl--dependent co-transport systems. Organic anions (e.g., PAH) undergo net secretion. This process involves entry into the cells at the basolateral membrane against an electrochemical gradient via a tertiary active process, the final step involving countertransport for a-ketoglutarate. Exit at the luminal membrane is down an electrochemical gradient, probably via anion exchange or carrier-mediated diffusion. Urate undergoes net secretion. This process involves transport into the cells against an electrochemical gradient via an unknown, Na+-independent, K+-sensitive, mechanism that may involve anion exchange and exit across the luminal membrane down an electrochemical gradient via what appears to be simple diffusion. Some organic cations, such as tetraethylammonium (TEA), undergo net secretion and some, such as N-1-methylnicotinamide (NMN), undergo net reabsorption. Net TEA secretion involves entry across the basolateral membrane, down an electrochemical gradient via carrier-mediated diffusion or organic anion exchange, and exit at the luminal membrane against an electrochemical gradient via countertransport for H+. Net NMN reabsorption involves entry across the luminal membrane, down an electrochemical gradient via Na+-dependent, carrier-mediated diffusion, with exit across the basolateral membrane against an electrochemical gradient via an unknown process that probably involves countertransport for an organic cation. (18.8-71.5 cm snout-vent length (SVL)) from the Lake Chad region in Chad and Nigeria and the central delta of the Niger River in Mali. The timing of egg maturation suggests that only one large clutch can be produced in 12 months, and that egg laying occurs between late November and early January. Each year, about 45-50% of mature females, on average, lay eggs, but local sampling shows that this proportion varies greatly. Females reach sexual maturity at 36 +/- 2 cm SVL and 24 months of age. Clutch size (CS) increases from 12-13 to 53-60 eggs (average values for specimens of 36 cm and 71.5 cm SVL, respectively) with increase in SVL, according to the power function CS = 0.0049 SVL2.206 (specimens from Mali) or CS = 0.0052 SVL2.163 (specimens from the Lake Chad region). Conversely, the relative mass of the clutches (RCM) and their relative energy content (RWC) decrease during growth; hence, small (young) individuals are subjected to a much higher reproductive effort than large ones. Compared with that of most other large varanids, egg mass in the Nile monitor (38.8 g) is small, but reproductive output (CS) and effort (RCM, RWC) appear to be very high. Moreover, the specimens from Mali, where the species is heavily exploited, have a higher reproductive output and shorter longevity than those from the Lake Chad region.
